Background of IL-15R alpha
Interleukin 15 receptor, alpha subunit is a subunit of the interleukin 15 receptor which is encoded by the gene IL15RA in humans. The human IL15RA gene was mapped to chromosome 10p15-p14 by FISH and the mouse Il15ra gene was mapped to chromosome 2. The cytoplasmic domain of IL15R-alpha, like that of IL2R-alpha, was dispensable for mitogenic signaling, suggesting that the primary role of the alpha chains is to confer high-affinity binding. At high concentrations, IL15, like IL2, was able to signal through a complex of IL2R-beta and -gamma in the absence of the alpha subunit. The soluble receptor displayed high affinity for IL15 and inhibited both binding of IL15 to the membrane receptor and IL15-induced cell proliferation.

Q: which procedure should I follow in order to thaw whole blood sample for IL-15R alpha ELISA after freezing?

Answer
A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test IL-15R alpha for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.

If sterile material is required for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.

A: Boster Bio does not assess sample stability. Variations in sample collection, processing, and storage may alter the stabilityof samples. It is recommend to assay sample on-stat collection when possible, or aliquot into single use volumes and store samples frozen. avoid repeat freeze-thaw cycles with the stored samples to avoid protein degradation.

A: in those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are included in the product insert. Components in lysate and lysis buffer may impact immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

Q: if the enzyme conjugated IL-15R alpha antibodies are mixed with the substrate, will that change the substrate into the enzymatic reaction product? Or the enzyme function is only activated when the antibody is attached with the IL-15R alpha antigen?
