Background of Clusterin/APOJ

Clusterin (apolipoprotein J) is a 75 - 80 kDa disulfide-linked heterodimeric protein associated with the clearance of cellular debris and apoptosis.[1] In humans, clusterin is encoded by the CLU gene.This protein has several synonyms: dimeric acidic glycoprotein (DAG protein), testosterone repressed prostate message-2 (TRPM-2), sulfated glycoprotein-2 (SGP-2) and complement lysis inhibitor (CLI). Clusterin was mapped to mouse chromosome 14. Clusterin is a ubiquitously expressed molecule thought to influence a variety of processes including cell death. In the brain, it accumulates in dying neurons following seizures and hypoxic-ischemic (H-I) injury. clusterin may be a new therapeutic target to modulate non-caspase-dependent neuronal death following acute brain injury.

The range of clusterin in saliva is 82-695ng/ml. Recommended diulution factor is around 1:20. The assay range of standard in the Human Clusterin ELISA Kit PicoKine™ (EK0914) is 0.78ng/ml-50ng/ml. Please estimate the concentration of the target protein in the sample and use an appropriate dilution factor so that the diluted target protein concentration falls in the middle range of O.D. values of the standard curve.

A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test Clusterin for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.

A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may change the stabilityof samples. It is recommend to assay sample on-stat collection when possible, or aliquot into single use volumes and store samples frozen. limit repetitive freeze-thaw cycles with the stored samples in order to limit protein degradation.
