The WAC Protein binds phosphoryler5 in a weak, but significant, manner. This protein may not play a significant role in recruiting RNF20/40/WAC genes to transcribed genes. Moreover, the WAC protein interacts with phospho-Ser5 of Pol II, and may not play a crucial role in recruiting RNF20/40/WAC to transcribed genes.
The RNF20/40 transcription factor interacts with WAC. WAC is a transcription element that regulates the ubiquitination and ubiquitination H2B. WAC interacts with RNF20/40 through its coiled-coil motif. WAC recognizes RNA polymerase II, a protein that is essential for ubiquitination, through its Nterminal WW domain. WAC-dependent transcription is also required for cell cycle activation.
Another study of the RNF20 gene found that the promoter of WAC and SFB-DWW mutants were over-expressed in breast cancer cells. These studies also revealed that RNF20 is expressed in tumor cells and normal cells. This study also demonstrated that RNF20 and WAC can interact with each other, suggesting that the downregulation of RNF20 inhibits tumor formation.
