E3 ubiquitin-protein ligase BRE1A (RNF20)

Part of the RNF20/40 E3 ubiquitin-protein ligase complex that mediates monoubiquitination of'Lys-120' of histone H2B (H2BK120ub1). H2BK120ub1 gives a specific tag for epigenetic transcriptional activation and is also prerequisite for histone H3 'Lys-4' and 'Lys-79' methylation (H3K4me and H3K79me, respectively).

It thereby plays a central role inb histone code and gene regulation. The RNF20/40 complex forms a H2B ubiquitin ligase complex in cooperation with the E2 enzyme UBE2A or UBE2B; reports about the cooperation with UBE2E1/UBCH are contradictory.

Boster Bio

Boster Bio is a major RNAi target with the RNF20 gene. The RNF20 protein is a key regulator in cell viability. It is also associated to WAC. WAC facilitates the association of RNF20 with Pol II. RNF20 and WAC are constitutively related proteins. They form a stable complex in vitro.

The WAC Protein binds phosphoryler5 in a weak, but significant, manner. This protein may not play a significant role in recruiting RNF20/40/WAC genes to transcribed genes. Moreover, the WAC protein interacts with phospho-Ser5 of Pol II, and may not play a crucial role in recruiting RNF20/40/WAC to transcribed genes.

The RNF20/40 transcription factor interacts with WAC. WAC is a transcription element that regulates the ubiquitination and ubiquitination H2B. WAC interacts with RNF20/40 through its coiled-coil motif. WAC recognizes RNA polymerase II, a protein that is essential for ubiquitination, through its Nterminal WW domain. WAC-dependent transcription is also required for cell cycle activation.

Another study of the RNF20 gene found that the promoter of WAC and SFB-DWW mutants were over-expressed in breast cancer cells. These studies also revealed that RNF20 is expressed in tumor cells and normal cells. This study also demonstrated that RNF20 and WAC can interact with each other, suggesting that the downregulation of RNF20 inhibits tumor formation.
