CISD2 is a transcription element that activates AKT. CISD2 phosphorylates AKT at Thr308 and Ser473. This protein is a known transcription activator. CISD2's effects are abated by inhibition of AKT kinase activity. Inhibition of CISD2's activity also reduces GSK3b, a protein implicated in the CISD2 pathway.
CISD2 interacts with GIMAP5 on mitochondria-associated ER membranes. It is necessary to maintain mitochondrial calcium homeostasis. We used primary antibodies of Sigma-Aldrich for this study. These antibodies were anti CISD2, beclin-1, LC3II (Atg7) autophagy related 7 (Atg7) anti-p62 and anti-rabbit anti-rabbit IgG. Recent studies have identified CISD2 a target for cancer therapy. The target gene CKEV contains a sequence known as KKEV. Furthermore, the CKEV sequence found on the Cterminus (C-terminus) of Cisd2 acts as an ER-targeting element. The C-terminal end of the gene does NOT colocalize to the Golgi marker. A small amount of the CKEV protein can be detected on the cell's surface.
