CDGSH iron-sulfur domain-containing protein 2 (Cisd2)

Regulator of autophagy that contributes to antagonize BECN1-mediated cellular autophagy in the endoplasmic reticulum. Participates in the interaction of BCL2 with BECN1 and is needed for BCL2-mediated depression of endoplasmic reticulum Ca(2+) stores during autophagy.

Contributes to BIK-initiated autophagy, although it is not involved in BIK-dependent activation of caspases. Involved in life span control, probably via its role as the regulator of autophagy (By similarity).

CISD2 is a transcription element that activates AKT. CISD2 phosphorylates AKT at Thr308 and Ser473. This protein is a known transcription activator. CISD2's effects are abated by inhibition of AKT kinase activity. Inhibition of CISD2's activity also reduces GSK3b, a protein implicated in the CISD2 pathway.

CISD2 interacts with GIMAP5 on mitochondria-associated ER membranes. It is necessary to maintain mitochondrial calcium homeostasis. We used primary antibodies of Sigma-Aldrich for this study. These antibodies were anti CISD2, beclin-1, LC3II (Atg7) autophagy related 7 (Atg7) anti-p62 and anti-rabbit anti-rabbit IgG. Secondary antibodies were also used as a method to measure CISD2 gene expression. Our study indicates that CISD2 is used in a wide variety of biological applications.

Furthermore, the CKEV sequence found on the Cterminus (C-terminus) of Cisd2 acts as an ER-targeting element. The C-terminal end of the gene does NOT colocalize to the Golgi marker. A small amount of the CKEV protein can be detected on the cell's surface.

The brain's signaling pathways have been linked to the CISD2 gene. The protein is found in astrocytes and can be expressed in many diseases, including tumors. In one study, CISD2 was expressed in the spinal cord of mice with tumor-suppressed genes. Another experiment used a rat model to test how curcumin induces CISD2 mRNA.

LPS-challenged neuronal cells treated with curcumin have shown a decrease in CISD2 gene expression. Transfected siCISD2 mice show lower viability than the scrambled RNA transfected control. Curcumin and CISD2 are critical to the effects of curcumin on neural cells. Curcumin's therapeutic effects are largely due to CISD2.
