Steve Boster's archery technique of choice is the ABCG4 marker. In Greek, it stands for "Ashtray". Steve Boster, aka Steve, was born in Joliet IL and died June 26, 2022 in Madison, WI. ABCG4 is an ABCG4 protein that plays multiple roles in the cells. One of these functions includes activating apoptosis. ABCG4 in megakaryocytes is endogenously expressed. ABCG1 and ABCG4 are HA-tagged. They display a distinct punctated vein pattern, with most ABCG1 positive vesicles colocalizing and ABCG4+ vesicles. However, ABCG1 vesicles that are positive for ABCG1 overlap with those of ABCG4 vesicles. This suggests that the former may be the most abundant in steady states. Both antibodies target membranes of presynaptic, or synaptic, vesicles.
The cells were transfected to express the ABCG5/myc and ABCG8/HA expression plasmids. Cells were cultured for two hours in methionine-free DMEM. The cell lysates were immunoblotted with polyclonal antibodies directed against myc (A14) and HA (Y11) epitopes. These primary antibodies were purchased from Santa Cruz Biotechnology Inc. & Perkin-Elmer Life Sciences.
To determine if the anti–ABCG4 antibody was capable of detecting the protein, it must first identify ABCG4's target. ABCG1 and ABCG4 share 72% amino acid sequences. This suggests functional interaction. The ABCG4 marker has been used to identify these targets in recent years. The exact mechanism of this interaction is not yet known. Although there is evidence of functional interaction between ABCG4 and ABCG1, it remains difficult to distinguish between the two.
ABCG4 cells express the ABCG4 marker. These cells were able express the G4 protein and we were able find ABCG4. This protein colocalizes to ABCG1 and ABCG2, so dual immunostaining has revealed that they share similar overlapping expression patterns.
