Background of Zp1
Zona pellucida sperm-binding protein 1 (ZP1) is a protein that belopngs to the mammalian zona pellucida. The zona pellucida is an extracellular matrix that surrounds the oocyte and early embryo. It is composed primarily of three or four glycoproteins with various functions during fertilization and preimplantation development. Zp1 ensures the structural integrity of the zona pellucida. Mutations in this gene are a cause of oocyte maturation defect and infertility. And this gene is mapped to chromosome 11q12.2.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: rat ovary tissue lysates,
Lane 2: SKOV3 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-ZP1 antigen affinity purified polyclonal antibody (Catalog # PB9970) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for ZP1 at approximately 70 kDa, 100 kDa. The expected band size for ZP1 is at 70 kDa.

We are currently using anti-ZP1 antibody PB9970 for rat tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, rat. Is it possible that the antibody can work on canine tissues as well?

The anti-ZP1 antibody (PB9970) has not been validated for cross reactivity specifically with canine tissues, but there is a good chance of cross reactivity.
