Background of TIF1 alpha
Transcriptional coactivator that interacts with numerous nuclear receptors and coactivators and modulates the transcription of target genes. Interacts with chromatin depending on histone H3 modifications, having the highest affinity for histone H3 that is both unmodified at 'Lys-4' (H3K4me0) and acetylated at 'Lys-23' (H3K23ac). Has E3 protein-ubiquitin ligase activity. Promotes ubiquitination and proteasomal degradation of p53/TP53. Plays a role in the regulation of cell proliferation and apoptosis, at least in part via its effects on p53/TP53 levels. Up-regulates ligand-dependent transcription activation by AR, GCR/NR3C1, thyroid hormone receptor (TR) and ESR1. Modulates transcription activation by retinoic acid (RA) receptors, including RARA. Plays a role in regulating retinoic acid-dependent proliferation of hepatocytes By similarity.

Does A03258-1 anti-TRIM24/Tif1 Alpha antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer
As indicated on the product datasheet, A03258-1 anti-TRIM24/Tif1 Alpha antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-TRIM24/Tif1 Alpha antibody A03258-1 for mouse tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse. Is it likely that the antibody can work on horse tissues as well?

The anti-TRIM24/Tif1 Alpha antibody (A03258-1) has not been tested for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity. Our lab technicians have not tested anti-TRIM24/Tif1 Alpha antibody A03258-1 on feline. You can run a BLAST between feline and the immunogen sequence of anti-TRIM24/Tif1 Alpha antibody A03258-1 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated feline samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in feline liver in WB, you can get your next antibody for free.
