Background of TLR3
TLR3 (Toll-like receptor 3) also known as CD283 (cluster of differentiation 283) is a protein that in humans is encoded by the TLR3 gene. The TLR3 gene is mapped to chromosome 4q35 by fluorescence in situ hybridization. TLR3 is a member of the Toll-like receptor family of pattern recognition receptors of the innate immune system. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This receptor is most abundantly expressed in placenta and pancreas, and is restricted to the dendritic subpopulation of the leukocytes. It recognizes dsRNA associated with viral infection, and induces the activation of NF-κB and the production of type I interferons. It may thus play a role in host defense against viruses. TLR3 recognizes double-stranded RNA, a form of genetic information carried by some viruses such as retroviruses. Upon recognition, TLR3 induces the activation of NF-κB to increase production of type I interferons which signal other cells to increase their antiviral defenses. Double-stranded RNA is also recognised by the cytoplasmic receptors RIG-I and MDA-5. TLR3 displays a protective role in mouse models of atherosclerosis.[4] In addition, TLR3 activators show effects on human vascular cells.

Figure 1. Western blot analysis of TLR3 using anti-TLR3 antibody (A00197).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Jurkat whole cell lysates,
Lane 2: mouse Raw264.7 whole cell lysates,
Lane 3: human K562 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TLR3 antigen affinity purified polyclonal antibody (Catalog # A00197) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TLR3 at approximately 120 kDa. The expected band size for TLR3 is at 104 kDa.

Question
Can you help my question with product A00197, anti-TLR3 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
K. Wu
Verified customer
Asked: 2017-05-25
Answer
We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A00197 anti-TLR3 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

Question
Does A00197 anti-TLR3 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
B. Edwards
Verified customer
Asked: 2013-03-06
Answer
As indicated on the product datasheet, A00197 anti-TLR3 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
