Background of NHE1/SLC9A1
NHE1, also known as SLC9A1, is an isoform of sodium–hydrogen antiporter that in humans is encoded by the SLC9A1 gene. It is mapped to 1p36.11. NHE1 is a ubiquitous membrane-bound enzyme involved in pH regulation of vertebrate cells. It is specifically inhibited by the diuretic drug amiloride and activated by a variety of signals including growth factors, mitogens, neurotransmitters, tumor promoters, and others. The plasma membrane ion exchanger NHE1 acts as an anchor for actin filaments to control the integrity of the cortical cytoskeleton. And a structural role for NHE1 in regulating the cortical cytoskeleton that is independent of its function as an ion exchanger. NHE1 is a plausible candidate gene for human essential hypertension.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: recombinant human Sodium protein 0.5 ng.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SLC9A1 antigen affinity purified polyclonal antibody (Catalog # PB9151) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for SLC9A1 at approximately 37 kDa. The expected band size for SLC9A1 is at 37 kDa.

Lane 1: rat PC-12 whole cell lysates,
Lane 2: human MCF-7 whole cell lysates.

The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 μg/ml rabbit anti-SLC9A1 Antibody (PB9151) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB as the chromogen.

We are currently using anti-Sodium/Hydrogen Exchanger 1/SLC9A1 antibody PB9151 for mouse tissue, and we are happy with the IHC results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on pig tissues as well?

The anti-Sodium/Hydrogen Exchanger 1/SLC9A1 antibody (PB9151) has not been validated for cross reactivity specifically with pig tissues, though there is a good chance of cross reactivity. As indicated on the product datasheet, PB9151 anti-Sodium/Hydrogen Exchanger 1/SLC9A1 antibody as been validated on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
