Background of RANK/TNFRSF11A
Receptor Activator of Nuclear Factor κ B (RANK), also known as TRANCE Receptor, is a type I membrane protein that is expressed on the surface of osteoclasts and is involved in their activation upon ligand binding. RANK is also expressed on dendritic cells and facilitates immune signaling.RANKL (Receptor Activator for Nuclear Factor κ B Ligand) is found on the surface of stromal cells, osteoblasts, and T cells. By analysis of somatic cell and radiation hybrid panels, Anderson et al. (1997) mapped the RANK gene to 18q22.1.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: HEPA Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-RANK antigen affinity purified polyclonal antibody (Catalog # PB9413) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for RANK at approximately 100 kDa. The expected band size for RANK is at 66 kDa.

Question
We are currently using anti-RANK/TNFRSF11A antibody PB9413 for mouse tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says mouse. Is it likely that the antibody can work on bovine tissues as well?

The anti-RANK/TNFRSF11A antibody (PB9413) has not been validated for cross reactivity specifically with bovine tissues, though there is a good chance of cross reactivity.
