Background of PTEN
This gene (PTENP1) is a highly homologous pseudogene of PTEN. PTEN was identified as a tumor suppressor that is mutated in a large number of cancers at high frequency. PTEN is a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase. It contains a tension like domain as well as a catalytic domain similar to that of the dual specificity protein tyrosine phosphatases. Unlike most of the protein tyrosine phosphatases, this protein preferentially dephosphorylates phosphoinositide substrates. It negatively regulates intracellular levels of phosphatidylinositol-3,4,5-trisphosphate in cells and functions as a tumor suppressor by negatively regulating AKT/PKB signaling pathway.

Western blot analysis of PTEN-P1 expression in human HeLa whole cell lysates (lane 4), HEK293 whole cell lysates (lane 5) and MCF7 whole cell lysates (lane 6). Lanes 1-3 were show the results of staining after the antibody was first pre-incubated with the immunizing peptide. The identity of lower molecular weight bands in lane 4 is unknown. Molecular weight estimation was made by comparison to prestained MW markers in lane M (700 nm channel, red). PTEN-P1 at 55KD was detected using rabbit anti-PTEN-P1 Antigen Affinity purified polyclonal antibody (Catalog # A08380) at 1:500. The blot was developed using chemiluminescence (ECL) method (Catalog # EK1002).

Question
My question regarding product A08380, anti-PTEN-P1 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We do not recommend storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A08380 anti-PTEN-P1 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

Our lab technicians have not tested anti-PTEN-P1 antibody A08380 on pig. You can run a BLAST between pig and the immunogen sequence of anti-PTEN-P1 antibody A08380 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated pig samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in pig cerebellar hemisphere in WB, you can get your next antibody for free.
