Background of FAT2
Recognizes a protein of 480kDa, which is identified as FAT2. The cadherins represent a family of Ca2+-dependent adhesion molecules that function to mediate cell-to-cell binding that is critical for the maintenance of structure and morphogenesis. Cadherins each contain a large extracellular domain at the N-terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short C-terminal intracellular domain interacts with a variety of cytoplasmic proteins, including -catenin, to regulate cadherin function. The cadherin superfamily includes cadherins, protocadherins, desmogleins and desmocollins. FAT2 (FAT tumor suppressor homolog 2) is a single-pass type I membrane protein that belongs to the protocadherin subfamily of cadherins. FAT2 contains one Laminin G-like domain, two EGF-like domains and 32 cadherin domains and is believed to function as a cell adhesion molecule, controlling cell proliferation and playing an important role in cerebellum development.

Assay Dilutions Recommendation

Flow Cytometry (1-2ug/million cells)
Immunofluorescence (1-2ug/ml)
Immunohistochemistry (Formalin-fixed) (1-2ug/ml for 30 min at RT)(Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at RT for 20 minutes)

Question
I have a question about product M04835, anti-Protocadherin FAT2 (FAT Atypical Cadherin 2) Monoclonal antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We do not advise storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M04835 anti-Protocadherin FAT2 (FAT Atypical Cadherin 2) Monoclonal antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

We are currently using anti-Protocadherin FAT2 (FAT Atypical Cadherin 2) Monoclonal antibody M04835 for human tissue, and we are happy with the IHC results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on horse tissues as well?

The anti-Protocadherin FAT2 (FAT Atypical Cadherin 2) Monoclonal antibody (M04835) has not been tested for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity.
