Background of Presenilin-2
Presenilin-2 is a protein that in humans is encoded by the PSEN2 gene. Kovacs et al. (1996) demonstrated that the expression patterns of PS1 and PS2 in the brain are extremely similar to each other and that messages for both are primarily detectable in neuronal populations. Immunochemical analyses indicated that PS1 and PS2 are similar in size and localize to similar intracellular compartments (endoplasmic reticulum and Golgi complex). Li et al. (1997) demonstrated that wildtype PS1 and PS2 are localized to the nuclear membrane, its associated interphase kinetochores, and the centrosomes.In melanocytic cells PSEN2 gene expression may be regulated by MITF.

Our lab technicians have not validated anti-Presenilin 2/PSEN2 antibody PA1358 on dog. You can run a BLAST between dog and the immunogen sequence of anti-Presenilin 2/PSEN2 antibody PA1358 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated dog samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in dog cervix carcinoma erythroleukemia in IHC, you can get your next antibody for free.

As indicated on the product datasheet, PA1358 anti-Presenilin 2/PSEN2 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-Presenilin 2/PSEN2 antibody PA1358 for mouse tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on feline tissues as well?

The anti-Presenilin 2/PSEN2 antibody (PA1358) has not been tested for cross reactivity specifically with feline tissues, but there is a good chance of cross reactivity.
