Background of Nicastrin
Nicastrin, also known as NCSTN, is a protein that is a part of the gamma secretase protein complex. It is mapped to 1q23.2. This gene is not catalytically active, but instead promotes the maturation and proper trafficking of the other proteins in the gamma secretase protein complex, all of which undergo significant post-translational modification before becoming active in the cell. NCSTN encodes a Type I transmembrane glycoprotein that is an integral component of the multimeric gamma-secretase complex. The encoded protein cleaves integral membrane proteins, including Notch receptors and beta-amyloid precursor protein, and may be a stabilizing cofactor required for gamma-secretase complex assembly. The cleavage of beta-amyloid precursor protein yields amyloid beta peptide, the main component of the neuritic plaque and the hallmark lesion in the brains of patients with Alzheimer's disease. Nicastrin has also been identified as a regulator of neprilysin, an enzyme involved in the degradation of amyloid beta fragment.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human 293T whole cell lysates,
Lane 2: human Hela whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-NCSTN antigen affinity purified polyclonal antibody (Catalog # PA2250) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for NCSTN at approximately 120-130 kDa. The expected band size for NCSTN is at 78 kDa.

Question
We are currently using anti-Nicastrin/NCSTN antibody PA2250 for rat tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on horse tissues as well?

The anti-Nicastrin/NCSTN antibody (PA2250) has not been validated for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity.
