Background of MRPS5
Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and help in protein synthesis within the mitochondrion. Mitochondrial ribosomes (mitoribosomes) consist of a small 28S subunit and a large 39S subunit. They have an estimated 75% protein to rRNA composition compared to prokaryotic ribosomes, where this ratio is reversed. Another difference between mammalian mitoribosomes and prokaryotic ribosomes is that the latter contain a 5S rRNA. Among different species, the proteins comprising the mitoribosome differ greatly in sequence, and sometimes in biochemical properties, which prevents easy recognition by sequence homology. This gene encodes a 28S subunit protein that belongs to the ribosomal protein S5P family. Pseudogenes corresponding to this gene are found on chromosomes 4q, 5q, and 18q.

Does anti-MRPS5 antibody A14183 work on horse IHC with cerebellar hemisphere?

Our lab technicians have not tested anti-MRPS5 antibody A14183 on horse. You can run a BLAST between horse and the immunogen sequence of anti-MRPS5 antibody A14183 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated horse samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in horse cerebellar hemisphere in IHC, you can get your next antibody for free.

Question
Would A14183 anti-MRPS5 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer
You can see on the product datasheet, A14183 anti-MRPS5 antibody as been validated on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

We are currently using anti-MRPS5 antibody A14183 for human tissue, and we are satisfied with the IHC results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on zebrafish tissues as well?
