Background of MATN3
MATN3 (Matrilin-3) is a protein that in humans is encoded by the MATN3 gene. This gene encodes a member of von Willebrand factor A domain containing protein family. This family of proteins is though to be involved in the formation of filamentous networks in the extracellular matrices of various tissues. By fluorescence in situ hybridization, Belluoccio et al. (1998) mapped the human MATN3 gene to chromosome 2p24-p23. Wagener et al. (2000) mapped the mouse Matn3 gene to the proximal end of chromosome 12, linked to the genes Synd1, Apob, Dtnb, and Kif3c. The human homologs of all 5 of these genes map to 2p23, indicating considerable homology of synteny. This protein contains two von Willebrand factor A domains; it is present in the cartilage extracellular matrix and has a role in the development and homeostasis of cartilage and bone. Mutations in this gene result in multiple epiphyseal dysplasia.

Our lab technicians have not validated anti-Matrilin 3/MATN3 antibody PA1927 on bovine. You can run a BLAST between bovine and the immunogen sequence of anti-Matrilin 3/MATN3 antibody PA1927 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated bovine samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in bovine cartilage in WB, you can get your next antibody for free.

Question
My question regarding product PA1927, anti-Matrilin 3/MATN3 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

It is not recommended storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free PA1927 anti-Matrilin 3/MATN3 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

We are currently using anti-Matrilin 3/MATN3 antibody PA1927 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on bovine tissues as well?
