Background of LCAT
LCAT (Lecithin: Cholesterol Acyltransferase), is an enzyme that converts free cholesterol into cholesteryl ester. Azoulay et al. (1987) used a cDNA clone corresponding to LCAT to assign the locus to 16q22 through the analysis of DNA from somatic cell hybrids and in situ hybridization. LCAT plays an important role in lipoprotein metabolism, especially in the process termed 'reverse cholesterol transport.' The enzyme is synthesized in the liver and circulates in blood plasma as a complex with components of high density lipoprotein (HDL). Cholesterol from peripheral cells is transferred to HDL particles, esterified through the action of LCAT on HDL, and incorporated into the core of the lipoprotein. The cholesterol ester is thereby transported to the liver (Jonas, 2000).

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Brain Tissue Lysate at 50ug,
Lane 2: Mouse Testis Tissue Lysate at 50ug,
Lane 3: HEPG2 Whole Cell Lysate at 40ug,
Lane 4: HELA Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-LCAT antigen affinity purified polyclonal antibody (Catalog # PB9657) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for LCAT at approximately 50 kDa. The expected band size for LCAT is at 50 kDa.

We are currently using anti-LCAT antibody PB9657 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on primate tissues as well?

The anti-LCAT antibody (PB9657) has not been validated for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity.
