Background of JAB1
COP9 constitutive photomorphogenic homolog subunit 5 (Arabidopsis), also known as COPS5 or JAB1, is a gene conserved from humans to Saccharomyces cerevisiae. It is a member of the MOV34 family. COPS5 is mapped to 8q13.1. The protein encoded by this gene is one of the eight subunits of COP9 signalosome, a highly conserved protein complex that functions as an important regulator in multiple signaling pathways. COPS5 can interact with the cytoplasmic domain of the beta-2 subunit of the alpha-L/beta-2 integrin LFA1, and it is the only protein demonstrated to interact with MIF. COPS5, VHL, and TRC8 proteins appear to be linked both physically and functionally, and all 3 may participate in the development of kidney cancer. In addition to that, COPS5 is an essential cofactor for the apoptotic function of E2F1.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Raji whole cell lysates,
Lane 2: human K562 whole cell lysates,
Lane 3: human PC-3 whole cell lysates,
Lane 4: human Hela whole cell lysates,
Lane 5: human A549 whole cell lysates,
Lane 6: human HEK293 whole cell lysates,
Lane 7: rat brain tissue lysates,
Lane 8: rat heart tissue lysates,
Lane 9: mouse brain tissue lysates,
Lane 10: mouse heart tissue lysates,
Lane 11: mouse SP2/0 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-JAB1 antigen affinity purified polyclonal antibody (Catalog # PB9490) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for JAB1 at approximately 38 kDa. The expected band size for JAB1 is at 38 kDa.

JAB1 was detected in a paraffin-embedded section of mouse testis tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1 μg/ml rabbit anti-JAB1 Antibody (PB9490) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB as the chromogen.

JAB1 was detected in immunocytochemical section of A431 cell. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent (AR0022) for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 1μg/ml rabbit anti-JAB1 Antibody (PB9490) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

Question
We are currently using anti-JAB1/COPS5 antibody PB9490 for mouse tissue, and we are content with the IF results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on pig tissues as well?

The anti-JAB1/COPS5 antibody (PB9490) has not been tested for cross reactivity specifically with pig tissues, but there is a good chance of cross reactivity.
