Background of IL32
IL-32A (also known as Natural killer cells protein 4, Tumor necrosis factor alpha-inducing factor, IL32α, Interleukin-32 and IL-32 isoform 4) is a member of the cytokine family. IL-32a is a secreted protein selectively expressed in lymphocytes and plays a role in innate and adaptive immune responses. The protein contains a tyrosine sulfation site, 3 potential N-myristoylation sites, multiple putative phosphorylation sites, and an RGD cell-attachment sequence. Expression of this protein is increased after the activation of T-cells by mitogens or the activation of NK cells by IL-2. This protein induces the production of TNFα and IL-8. It induces typical cytokine pathways of NF-kB and p38 MAPK. Alternate transcriptional splice variants, encoding different isoforms, have been characterized.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-IL32 antigen affinity purified polyclonal antibody (Catalog # A03286-1) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-Rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # SA1022) with Tanon 5200 system. A specific band was detected for IL32.
