Background of GFI-1
Zinc finger protein Gfi-1 is a protein that in humans is encoded by the GFI1 gene. This gene encodes a nuclear zinc finger protein that functions as a transcriptional repressor. This protein plays a role in diverse developmental contexts, including hematopoiesis and oncogenesis. It functions as part of a complex along with other cofactors to control histone modifications that lead to silencing of the target gene promoters. Mutations in this gene cause autosomal dominant severe congenital neutropenia, and also dominant nonimmune chronic idiopathic neutropenia of adults, which are heterogeneous hematopoietic disorders that cause predispositions to leukemias and infections. Multiple alternatively spliced variants, encoding the same protein, have been identified for this gene.

GFI1 was detected in paraffin-embedded section of human mammary cancer tissue. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/ml rabbit anti-GFI1 Antibody (A00888-1) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

We are currently using anti-GFI1 antibody A00888-1 for rat tissue, and we are content with the IHC results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on primate tissues as well?

The anti-GFI1 antibody (A00888-1) has not been validated for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity.
