Background of GATA-5
Transcription factor GATA-5, also named GATAS or GATA5, is a protein that in humans is encoded by the GATA5 gene. It is mapped to 20q13.33. The protein encoded by this gene is a transcription factor that contains two GATA-type zinc fingers. The encoded protein is known to bind to hepatocyte nuclear factor-1alpha (HNF-1alpha), and this interaction is essential for cooperative activation of the intestinal lactase-phlorizin hydrolase promoter. In other organisms, similar proteins may be involved in the establishment of cardiac smooth muscle cell diversity. In addition, the expression of GATA5 increased in a human colon cancer cell line within 2 hours of exposure to sodium butyrate, which induces differentiation.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human 293T whole cell lysates,
Lane 2: human HepG2 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GATA5 antigen affinity purified polyclonal antibody (Catalog # PB9200) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for GATA5 at approximately 50 kDa. The expected band size for GATA5 is at 41 kDa.

We are currently using anti-GATA5 antibody PB9200 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human, rat. Is it true that the antibody can work on feline tissues as well?

The anti-GATA5 antibody (PB9200) has not been validated for cross reactivity specifically with feline tissues, but there is a good chance of cross reactivity.
