Background of GABA-A R gamma 2

Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system. There are two major classes of GABA receptors: the GABA-A and the GABA-AB subtype of receptors. GABA-A-Rs are important therapeutic targets for a range of sedative, anxiolytic, and hypnotic agents and are implicated in several diseases including epilepsy, anxiety, depression, and substance abuse. The GABA-A-R is a multimeric subunit complex. To date six αs, four βs and four γs, plus alternative splicing variants of some of these subunits, have been identified (Olsen and Tobin, 1990; Whiting et al., 1999; Ogris et al., 2004). Injection in oocytes or mammalian cell lines of cRNA coding for α- and β-subunits results in the expression of functional GABA-A-Rs sensitive to GABA. However, coexpression of a γ-subunit is required for benzodiazepine modulation. It has recently been suggested that PKCε regulates the sensitivity of GABA-A α1β2γ2 receptors to ethanol and benzodiazepines through phosphorylation of serine 327 in the large intracellular loop of γ2 (Qi et al., 2007)

Question

Can you help my question with product P02361, anti-Phospho-GABAA Receptor gamma2 (Ser327) antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We do not advise storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free P02361 anti-Phospho-GABAA Receptor gamma2 (Ser327) antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

Our lab technicians have not validated anti-Phospho-GABAA Receptor gamma2 (Ser327) antibody P02361 on primate. You can run a BLAST between primate and the immunogen sequence of anti-Phospho-GABAA Receptor gamma2 (Ser327) antibody P02361 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated primate samples, this use of the antibody is not covered by our guarantee.
