Background of Connexin 43/GJA1
Gap junctional intercellular communication is thought to play a key role in development and may also be involved in epilepsy (Aronica et al., 2001). Connexin43 forms gap-junctional channels and regulates the permeability of these gap junctions to small organic molecules. Permeability of connexin43 is known to be regulated by phosphorylation at Ser-368 by protein kinase C (Yogo et al., 2002; Bao et al., 2004a). Phosphorylation of Ser-368 by PKC induces a conformational change of connexin43 that results in a decrease in gap junction permeability (Bao et al., 2004b).

My boss were content with the WB result of your anti-Phospho-Connexin43 (Ser368) antibody. However we have seen positive staining in pigmented layer of retina cell membrane using this antibody. Is that expected? Could you tell me where is GJA1 supposed to be expressed?

Based on literature, pigmented layer of retina does express GJA1. Generally GJA1 expresses in cell membrane. Regarding which tissues have GJA1 expression, here are a few articles citing expression in various tissues:

We are currently using anti-Phospho-Connexin43 (Ser368) antibody P00599 for mouse tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says mouse, rat. Is it possible that the antibody can work on monkey tissues as well?

The anti-Phospho-Connexin43 (Ser368) antibody (P00599) has not been tested for cross reactivity specifically with monkey tissues, though there is a good chance of cross reactivity.
