Background of CHX10
Visual system homeobox 2 is a protein that in humans is encoded by the VSX2 gene. This gene encodes a homeobox protein originally described as a retina-specific transcription factor. Mutations in this gene are associated with microphthalmia, cataracts and iris abnormalities. In situ hybridization to human fetal retinal sections detected CHX10 expression in retinal neuroblasts at all stages examined. Human CHX10 is expressed in progenitor cells of the developing neuroretina and in the inner nuclear layer of the mature retina.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: mouse eye ball tissue lysate.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CHX10 antigen affinity purified polyclonal antibody (Catalog # A05180) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CHX10 at approximately 43KD. The expected band size for CHX10 is at 39KD.

We are currently using anti-CHX10/VSX2 antibody A05180 for mouse tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse. Is it likely that the antibody can work on primate tissues as well?

The anti-CHX10/VSX2 antibody (A05180) has not been tested for cross reactivity specifically with primate tissues, but there is a good chance of cross reactivity.
