Background of CCL19/MIP-3 beta
Chemokine (C-C motif) ligand 19 (CCL19) is a protein that in humans is encoded by the CCL19 gene. This gene is one of several CC cytokine genes clustered on the p-arm of chromosome 9. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene may play a role in normal lymphocyte recirculation and homing. It also plays an important role in trafficking of T cells in thymus, and in T cell and B cell migration to secondary lymphoid organs. It specifically binds to chemokine receptor CCR7.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: rat thymus tissue lysate,
Lane 2: rat lung tissue lysate,
Lane 3: rat spleen tissue lysate,
Lane 4: rat stomach tissue lysate,
Lane 5: rat PC-12 whole Cell lysate,
Lane 6: mouse thymus tissue lysate,
Lane 7: mouse NIH3T3 whole Cell lysate.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Ccl19/MIP-3 beta antigen affinity purified polyclonal antibody (Catalog # A01605-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Ccl19/MIP-3 beta at approximately 20KD. The expected band size for Ccl19/MIP-3 beta is at 11KD.

We are currently using anti-Macrophage Inflammatory Protein 3 beta/Ccl19 antibody A01605-1 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says mouse, rat. Is it likely that the antibody can work on feline tissues as well?

The anti-Macrophage Inflammatory Protein 3 beta/Ccl19 antibody (A01605-1) has not been validated for cross reactivity specifically with feline tissues, but there is a good chance of cross reactivity.
