Background of Aquaporin-0
Lens fiber major intrinsic protein, also called MIP26 or MP26, is a protein that in humans is encoded by the MIP gene. MIP is a member of the water-transporting aquaporins as well as the original member of the MIP family of channel proteins. Using 2-color fluorescence in situ hybridization on high-resolution R-banded chromosomes and human genomic DNA clones for MIP as probes, this gene was found that located in close proximity in region 12q13. MIP plays a crucial role in the development of a transparent eye lens. This gene may be responsible for regulating the osmolarity of the lens and interactions between homotetramers from adjoining membranes may stabilize cell junctions in the eye lens core.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50 ug of sample under reducing conditions.
Lane 1: rat eye ball tissue Lysate.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Aquaporin 0 antigen affinity purified polyclonal antibody (Catalog # PB9811) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Aquaporin 0 at approximately 28 kDa. The expected band size for Aquaporin 0 is at 28 kDa.

We are currently using anti-Aquaporin 0/MIP antibody PB9811 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, rat. Is it likely that the antibody can work on monkey tissues as well?

The anti-Aquaporin 0/MIP antibody (PB9811) has not been tested for cross reactivity specifically with monkey tissues, though there is a good chance of cross reactivity.
