Background of alpha Tubulin
Alpha-tubulin (b-alpha-1) mRNA is expressed only in brain with a molecular weight of about 55,000. The 3-prime UTR of b-alpha-1 is more than 80% homologous to the UTR of the rat brain alpha-tubulin gene, IL-alpha-T1. B-alpha-1 encodes a predicted 451-amino acid protein that is 100% identical to the rat homolog and differs by only 2 and 3 amino acids from the pig and chicken homologs, respectively.

alpha-Tubulin was detected in a paraffin-embedded section of human mammary cancer tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1 μg/ml mouse anti-alpha-Tubulin Antibody (MA1107) overnight at 4°C. Peroxidase Conjugated Goat Anti-mouse IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Mouse IgG Super Vision Assay Kit (Catalog # SV0001) with DAB as the chromogen.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human SH-SY5Y whole cell lysates,
Lane 2: human Jurkat whole cell lysates,
Lane 3: human A549 whole cell lysates,
Lane 4: rat brain tissue lysates,
Lane 5: mouse brain tissue lysates,
Lane 6: mouse thymus tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with mouse anti-alpha-Tubulin antigen affinity purified monoclonal antibody (Catalog # MA1107) at 1 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-mouse IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1001) with Tanon 5200 system. A specific band was detected for alpha-Tubulin at approximately 55 kDa. The expected band size for alpha-Tubulin is at 50 kDa.

Our lab technicians have not tested anti-Alpha-Tubulin antibody (Monoclonal, DM1A) MA1107 on monkey. You can run a BLAST between monkey and the immunogen sequence of anti-Alpha-Tubulin antibody (Monoclonal, DM1A) MA1107 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated monkey samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in monkey brain in WB, you can get your next antibody for free.
