Background of TFF1/pS2
TFF1 (Trefoil factor 1), also known as pS2, is a protein that in humans is encoded by the TFF1 gene. Members of the trefoil family are characterized by having at least one copy of the trefoil motif, a 40-amino acid domain that contains three conserved disulfides. They are stable secretory proteins expressed in gastrointestinal mucosa. Their functions are not defined, but they may protect the mucosa from insults, stabilize the mucus layer, and affect healing of the epithelium. It is found that TFF1 in normal human urine inhibited the growth of calcium oxalate crystals. Urinary TFF1 showed an inhibitory potency similar to that of nephrocalcin, and inhibition was dose dependent and inhibited by TFF1 antisera, particularly by antisera directed to the TFF1 C terminus. Concentrations and relative amounts of TFF1 in the urine of patients with idiopathic calcium oxalate kidney stones were significantly less than those found in controls. This gene, which is expressed in the gastric mucosa, has also been studied because of its expression in human tumors. This gene and two other related trefoil family member genes are found in a cluster on chromosome 21.

"The TFF1 ELISA kit from Boster was competitively priced and worked with cell culture media taken from MCF7 cells, a human mammary carcinoma which is well known for overexpression of the TFF1 protein. The kit is very sensitive, detecting TFF1 protein at levels of 10pg/mL. The kit is saturated about 1mg/mL, so in my case, a 1 to 100 dilution of cell culture media was needed in order to be in the linear range of the assay. Good ELISA kit for human TFF1 protein. Worked as advertised."

The volume of Biotinylated antibody and Avidin-Biotin-Peroxidase Complex have been changed from 130ul to 100ul. All future lots contains 100 ul for those two components. Our lab takes reasonable loss into account and provide more than 100ul and they are sufficient for users to finish the assay.

A: Unfortunately, Boster Bio has not routinely validated tissue homogenates as a sample type for ELISA kits. This does not mean that ELISA kits are not valid for other sample types than we have tested: it means further investigation is required. One will need to perform a spike and recovery study to determine if an unvalidated sample type will work with a particular kit. To perform a spike and recovery experiment, one should divide a sample into two aliquots. In one of the aliquots, the user should spike in a known amount of the kit standard. a dilution series is performed comparing the spiked versus the unspiked sample. Generally, samples with expected recovery and linearity between 80-120% are considered acceptable. This method can be used to validate any sample type that has not been evaluated by Boster Bio. for a more detailed spike and recovery protocol, please contact technical support.
Note: acceptable ranges should be determined individually by each laboratory. Additionally, technical support can help determine if a buffer component is not compatible with a given ELISA kit. please check the Citations tab on the product webpage for peer-reviewed papers utilizing a wide range of sample types. A: for those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate can be found in the product insert. Components in lysate and lysis buffer can affect immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

A: without having an understanding the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the TFF1 ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.
