Background of IL-6
The human interferon-beta 2 gene (IFNB2) product is identical to that for the B-cell stimulation factor-2 (BSF-2), the hybridoma growth factor (HGF) ("interleukin-6"), and the hepatocyte stimulating factor (HSF). Proteins derived from this gene mediate the plasma protein response to tissue injury (acute-phase response) and regulate the growth and differentiation of both B and T cells. Interleukin-6 (IL6) has come to be regarded as a potential osteoporotic factor because it has stimulatory effects on cells of the osteoclast lineage, and, thus, may play a role in the pathogenesis of bone loss associated with estrogen deficiency. IL-6 has many roles essential to the regulation of the immune response, hematopoiesis, and bone resorption. It is involved not only in the hepatic acute phase response but also in adipose tissue metabolism, lipoprotein lipase activity, and hepatic triglyceride secretion. Overproduction of IL-6, a proinflammatory cytokine, is associated with a spectrum of age-related conditions including cardiovascular disease, osteoporosis, arthritis, type 2 diabetes, certain cancers, periodontal disease, frailty, and functional decline. BSF-2 is a novel interleukin consisting of 184 amino acids. The standard product used in this kit is recombinant human IL-6, consisting of 184 amino acids with the molecular mass of 20.3kDa.

The Human IL-6 ELISA Kit comes with 96 well with removable strips, so you can remove as many strips as you need before using it, then store the remaining strips in the pouch with desiccant, sealed it tightly and put in storage at 4C. For your data analysis excel will do. All you need to do is to calculate the average absorbance values and create a standard curve, then interpolate the sample concentrations from the standard curve.

A: Chelating agents such as EDTA, Heparin and Citrate can irreversibly bind metal ions from the functional domain of IL-6 causing disruption of its protein structure. IL-6 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes needed for detection and block the antigen antibody reaction. We have tested the IL-6 ELISA, treating samples with a number of anticoagulants and decided that heparin, EDTA or citrate can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

For the Human IL-6 ELISA Kit PicoKine™ (EK0410), it is normal that there is fluid in the wells of the plate, and the drops could be removed by patting the plate on a paper towel or by aspiration. For the standard, we never encountered this problem before, and our technicians suggests to centrifuge at 2000-4000×g for 5 minutes after dissolving the standard.

The Human IL-6 EK0410 Elisa Kit and Human IL-10 EK0416 Elisa Kit have not been tested on tissue homogenates. Endogenous biotin in tissue homogenates might introduce a false positive signal. We recommend to run a pilot test to confirm the effect of the endogenous biotin first. Add tissue homogenates into the wells and then add ABC and TMB without adding any biotinylated detection antibody to see if any signal will be observed. If no signal is produced, proceed and work on the tissue sample by using the kit.
