  • Deionized or sterile water
  • Squirt bottle, manifold dispenser, multichannel pipette reservoir or automated microplate washer
  • Graph paper or computer software capable of generating or displaying logarithmic functions

What is your protocol for EKC1161 based on serum test?

Answer

The following is the protocol for Cytochrome c Colorimetric Cell-Based ELISA Kit EKC1161 used on serum: 1) Collect whole blood into a tube without additives. 2) Keep the blood at room temperature for 30 min. 3) Centrifuge at 3,000 rpm and 4°C for 10 min. 4) Assay immediately or aliquot supernatant (serum) and hold at -80°C (Avoid freeze/thaw cycles). Note: Hemolysis should be avoided while collecting serum samples. Those samples that have undergone hemolysis may increase non-specifically staining in HRP-conjugated ELISA assay.

The Cytochrome C Colorimetric Cell-Based ELISA Kit (EKC1161) and Caspase 3 Colorimetric Cell-Based ELISA Kit (EKC1086) are specifically geared towards whole cells (adherent or suspension) seeded to the bottom of the plate. It is not intended to be used with cell lysates or any other types of samples.

Can frozen tissue extracts be used as a sample for EKC1086 and EKC1161? What should be the weight or volume of sample?

Our cell-based ELISA kits are meant to be used with cells being plated, grown, then fixed on the wells. They are not meant to be used with cell lysates or tissue homogenates, as they have not been validated for that purpose. We cannot offer any sort of guarantee using it on tissue samples.
