Research Spotlight - May 2022

For this month’s Research Spotlight, we highlight 10 new research publications from scientists whom Boster has served.

In the field of cancer immunotherapy, data from two studies point toward the anti-lymphoma and anti-bladder cancer effects exerted by ADAM10 and iDAMP inhibitors respectively. Additionally, scientists present the promising potential of CEACAM 5, PCL22-186, and lncSLCO1C1 as new biomarkers for predicting cancer cells’ radiosensitivity, cancer progression, and resistance.

Shedding new light on the current global pandemic, one research paper suggests ERK/VEGF as potential biomarkers and therapeutic targets for COVID-19 disease progression. Another researcher also discusses how sequential immunization of inactivated bivalent vaccines could be a potentially effective countermeasure to combat future COVID-19 variants.

Inhibitors of ADAM10 Reduce Hodgkin Lymphoma Cell Growth in 3D Microenvironments and Enhance Brentuximab-Vedotin Effect

Authors: Pece, R., Tavella, S., Costa, D., Varesano, S., Camodeca, C…
Journal: Haematologica

Shedding of ADAM10 substrates, like TNFa or CD30, can affect both anti-tumor immune response and antibody-drug-conjugate (ADC)-based immunotherapy. We have published two new ADAM10 inhibitors, LT4 and MN8 able to prevent such shedding in Hodgkin lymphoma (HL). Since tumor tissue architecture deeply influences the outcome of anti-cancer...

PCL22-186: A Potential Biomarker for Radiosensitivity of Squamous Cell Carcinomas in the Head and Neck

Authors: Bernardino, L., Kooiker, P., Garcia, J.C., Boudreaux, C., Bollag, R.
Journal: Journal of the National Comprehensive Cancer Network

Patients with squamous cell carcinoma in the head and neck often undergo radiation treatment with unwanted side effects and tumor recurrence. A biomarker that will predict response to radiotherapy will benefit these patients. Many tumors, including squamous cell carcinomas of the head and neck, express CEACAM 5. This gene encodes...

Analysis of Extracellular Vesicle DNA at the Single-Vesicle Level by Nano-Flow Cytometry

Authors: Liu, H., Tian, Y., Xue, C., Niu, Q., Chen, C., Yan, X.
Journal: Journal of Extracellular Vesicles

It has been demonstrated recently that extracellular vesicles (EVs) carry DNA; however, many fundamental features of DNA in EVs (EV-DNA) remain elusive. In this study, a laboratory-built nano-flow cytometer (nFCM) that can detect single EVs as small as 40 nm in diameter and single DNA fragments of 200 bp upon SYTO 16 staining was...

Sequential Immunizations Confer Cross-Protection Against Variants of SARS-CoV-2, Including Omicron in Rhesus Macaques

Authors: Deng, W., Lv, Q., Li, F., Liu, J., Song, Z., Qi, F., Wei, Q., Yu, P., Liu, M…
Journal: Signal Transduction and Targeted Therapy

Variants of concern (VOCs) like Delta and Omicron, harbor a high number of mutations, which aid these viruses in escaping a majority of known SARS-CoV-2 neutralizing antibodies (NAbs). In this study, Rhesus macaques immunized with 2-dose inactivated vaccines (Coronavac) were boosted with an additional dose of homologous vaccine or...

Cell Death-Induced Immunogenicity Enhances Chemoimmunotherapeutic Response by Converting Immune-Excluded Into T-Cell Inflamed Bladder Tumors

Authors: Nikolos, F., Hayashi, K., Hoi, X.P., Alonzo, M.E., Mo, Q., Kasabyan, A...
Journal: Nature Communications

Chemoimmunotherapy has recently failed to demonstrate significant clinical benefit in advanced bladder cancer patients; and the mechanism(s) underlying such suboptimal response remain elusive. To date, most studies have focused on tumor-intrinsic properties that render them “immune-excluded”. Here, we explore an alternative, drug

SARS-CoV-2 Spike Spurs Intestinal Inflammation via VEGF Production in Enterocytes

Authors: Zeng, F.M., Li, Y.W., Deng, Z.H., He, J.Z., Li, W., Wang, L., Lyu, T…
Journal: EMBO Molecular Medicine

Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) can cause gastrointestinal (GI) symptoms that often correlate with the severity of COVID-19. Here, we explored the pathogenesis underlying the intestinal inflammation in COVID-19. Plasma VEGF level was particularly elevated in patients with GI symptoms and significantly...

Role of lncSLCO1C1 in Gastric Cancer Progression and Resistance to Oxaliplatin Therapy

Authors: Xiao, Y.F., Li, B.S., Liu, J.J., Wang, S.M., Liu, J., Yang, H., Hu, Y.Y…
Journal: Clinical and Translational Medicine

Background: Gastric carcinoma (GC) is one of the most deadly diseases due to tumour metastasis and resistance to therapy. Understanding the molecular mechanism of tumour progression and drug resistance will improve therapeutic efficacy and develop novel intervention strategies. Methods: Differentially expressed long non-coding RNAs...

Neuron Derived Fractalkine Promotes Microglia to Absorb Hematoma via CD163/HO-1 After Intracerebral Hemorrhage

Authors: You, M., Long, C., Wan, Y., Guo, H., Shen, J., Li, M., He, Q., Hu, B.
Journal: Cellular and Molecular Life Sciences

Background: Hematoma leads to progressive neurological deficits and poor outcomes after intracerebral hemorrhage (ICH). Early clearance of hematoma is widely recognized as an essential treatment to limit the damage and improve the clinical prognosis. CD163, alias hemoglobin (Hb) scavenger receptor on microglia, plays a pivotal role in...

Hepatic Deficiency of Selenoprotein S Exacerbates Hepatic Steatosis and Insulin Resistance

Authors: Qiao, L., Men, L., Yu, S., Yao, J., Li, Y., Wang, M., Yu, Y., Wang, N…
Journal: Cell Death & Disease

Nonalcoholic fatty liver disease (NAFLD) is closely associated with insulin resistance (IR) and type 2 diabetes mellitus (T2DM), which are all complex metabolic disorders. Selenoprotein S (SelS) is an endoplasmic reticulum (ER) resident selenoprotein involved in regulating ER stress and has been found to participate in the occurrence...

Journal: Journal of Innate Immunity

As a serious and elusive syndrome caused by infection, sepsis causes a high rate of mortality around the world. Our investigation aims at exploring the role and possible mechanism of specificity protein-1 (SP1) in the development of sepsis. A mouse model of sepsis was established by cecal ligation perforation, and a cellular model was...
