Research Spotlight – June 2020

The June edition of Research Spotlight features 10 recent research discoveries from scientists whom Boster has served.

This month’s selected research publications investigate several cancers and diseases, including hepatocellular carcinoma, hepatitis B, prostate cancer, ankylosing spondylitis, rheumatoid arthritis, and diabetes.

Among the studies, scientists propose a prodrug for inflammatory diseases as well as a tumor nanovaccine that utilizes electrostatic interactions. The relationship of depression with inflammation and metabolic dysfunction in addition to ketamine’s potential anti-depressive effects are also explored.

A hydrogen peroxide responsive prodrug of Keap1-Nrf2 inhibitor for improving oral absorption and selective activation in inflammatory conditions

Authors: Lu, M., Zhang, X., Zhao, J., You, Q., & Jiang, Z.
Journal: Redox Biology

Transcription factor Nuclear factor erythroid 2-related factor 2 (Nrf2) and its negative regulator, the E3 ligase adaptor Kelch-like ECH-associated protein 1 (Keap1), control the redox and metabolic homeostasis and oxidative stress. Inhibitors of Keap1-Nrf2 interaction are promising in oxidative stress related inflammatory diseases but now hit hurdles. By utilizing...

The immunoglobulin superfamily member 3 (IGSF3) promotes hepatocellular carcinoma progression through activation of the NF-κB pathway

Authors: Sheng, P., Zhu, H., Zhang, W., Xu, Y., Peng, W., Sun, J., Gu, M., & Jiang, H.
Journal: Annals of Translational Medicine

Background: Patients with hepatocellular carcinoma (HCC) suffer from a high fatality rate, likely due to increased incidence of tumor relapse and metastasis. Understanding the molecular mechanisms that contribute to HCC development and progression is vital for the discovery of new treatment targets. This study aims to explore...

Characterization of IL-21-expressing recombinant hepatitis B virus (HBV) as a therapeutic agent targeting persisting HBV infection

Authors: Shen, Z., Wu, J., Gao, Z., Wang, J., Zhu, H., Mao, R., Wang, X., Zhang, J...
Journal: Theranostics

Chronic infection by hepatitis B virus (HBV) is associated with high risks of liver fibrosis, cirrhosis and hepatocellular carcinoma. In mouse models of HBV persistence, interleukin 21 (IL-21) has been identified as a potent inducer of viral clearance. Strict hepatotropism makes recombinant HBV (rHBV) vectors ideal for liver-targeting gene delivery...

Synergistic tumor immunological strategy by combining tumor nanovaccine with gene-mediated extracellular matrix scavenger

Authors: Hu, Y., Lin, L., Chen, J., Maruyama, A., Tian, H., & Chen, X.
Journal: Biomaterials

The antitumor efficacy of tumor vaccines is often limited by weak T cell responses and poor activated T cells infiltration. Herein, we reported a novel synergistic strategy to simultaneously overcome these two obstacles to realize enhanced tumor elimination. To induce the robust T cell responses, we designed a minimalist tumor nanovaccine based on stepwise...

Protein and gene markers of metabolic dysfunction and inflammation together associate with functional connectivity in reward and motor circuits in depression

Authors: Goldsmith, D.R., Bekhbat, M., Le, N.A., Chen, X., Woolwine, B.J., Li, Z...
Journal: Brain, Behavior, and Immunity

Bidirectional relationships between inflammation and metabolic dysfunction may contribute to the pathophysiology of psychiatric illnesses like depression. Metabolic disturbances drive inflammation, which in turn exacerbate metabolic outcomes including insulin resistance. Both inflammatory (e.g. endotoxin, vaccination) and...

LncRNA SNHG17 aggravated prostate cancer progression through regulating its homolog SNORA71B via a positive feedback loop

Authors: Wu, G., Hao, C., Qu, X., Nie, J., Zhou, W., Huang, J., & He, Q.
Journal: Cell Death & Disease

Prostate cancer (PC) is a prevalent male malignancy with high occurrence rate. Recent studies have showed that small nucleolar host genes (SNHGs) and their homolog small nucleolar RNAs (snoRNAs) elicit regulatory functions in carcinogenesis. Present study aimed to investigate the role of SNHG17 and its homolog SNORA71B in PC. Function of SNHG17...

Bone marrow mesenchymal stem cells improve bone erosion in collagen-induced arthritis by inhibiting osteoclasia-related factors and differentiating into chondrocytes

Authors: Gao, J., Zhang, G., Xu, K., Ma, D., Ren, L., Fan, J., Hou, J., Han, J...
Journal: Stem Cell Research & Therapy

Background: Rheumatoid arthritis (RA) is characterized by joint inflammation and damage to the cartilage and bone in collagen-induced arthritis (CIA). Mesenchymal stem cells (MSCs) can improve articular symptoms and reduce bone erosion in CIA rats; however, the underlying mechanism remains unknown. This study aimed to investigate the...

Down-regulated miR-495 can target programmed cell death 10 in ankylosing spondylitis

Authors: Ni, W.J. & Leng, X.M.
Journal: Molecular Medicine

Background: MicroRNAs (miRNAs) play crucial roles in regulating eukaryotic gene expression. Recent studies indicated that aberrantly expressed miRNAs are involved in the pathogenesis of ankylosing spondylitis (AS). Indeed, hsa-miR-495-3p (miR-495) has been reported as an anti-oncogene in different cancers. However, the role of miR-495 in AS is still unknown...

Authors: Wang, C., Xu, X., Chen, J., Kang, Y., Guo, J., Duscher, D., Yang, X...
Journal: Frontiers in Bioengineering and Biotechnology

Background: Diabetes mellitus is a worldwide disease with high incidence. Diabetic peripheral neuropathy (DPN) is one of the most common but often ignored complications of diabetes mellitus that cause numbness and pain, even paralysis. Recent studies demonstrate that Schwann cells (SCs) in the peripheral nervous system play an essential...
