Research Spotlight - January 2021

Our January edition of Research Spotlight presents an array of 10 interesting developments in the biomedical world. Valuable information about proliferation of cancer cells, immunity, inflammatory bowel diseases, and the body’s responses to oxidative stress and various nutrients are discussed.

In addition, scientists investigate how SLC19A1 as the major transporter of CDNs into cells could serve as a new potential immunotherapeutic treatment of cancer and some inflammatory diseases.

Scroll down to learn more about these new scientific discoveries.

Serum Vitamins D, B9 and B12 in Greek Patients with Inflammatory Bowel Diseases

Authors: Gioxari, A., Amerikanou, C., Papada, E., Zioga, E., Georgoulis, A.D…
Journal: Nutrients

Deficiencies in vitamin D, folate and cobalamin are common in Inflammatory Bowel Disease (IBD). The aim of the present study was to assess serum levels of these vitamins in IBD adults based on the respective serum cut off values for vitamin deficiencies, and to explore possible associations with IBD-related biomarkers and nutritional...

SLC19A1 Transports Immunoreactive Cyclic Dinucleotides

Authors: Luteijn, R.D., Zaver, S.A., Gown, B.G., Wyman, S.K., Garelis, N.E…
Journal : Nature

The accumulation of DNA in the cytosol serves as a key immunostimulatory signal associated with infections, cancer and genomic damage1,2. Cytosolic DNA triggers immune responses by activating the cyclic GMP–AMP synthase (cGAS)–stimulator of interferon genes (STING) pathway3. The binding of DNA to cGAS activates its enzymatic activity...

A Novel Long Non‐Coding RNA lnc030 Maintains Breast Cancer Stem Cell Stemness by Stabilizing SQLE mRNA and Increasing Cholesterol Synthesis

Authors : Qin, Y., Hou Y., Liu, S., Zhu, P., Wan, X., Zhao, M., Peng, M., Zeng, H…
Journal : Advanced Science

Cancer stem cells (CSCs) are considered the roots of cancer metastasis and recurrence (CSCs), due in part to their self‐renewal and therapy resistance properties. However, the underlying mechanisms for the regulation of CSC stemness are poorly understood. Recently, increasing evidence shows that long non‐coding RNAs (lncRNAs) are...

Wisp1 is a Circulating Factor that Stimulates Proliferation of Adult Mouse and Human Beta Cells

Authors: Fernandez-Ruiz, R., García-Alamán, A., Esteban, Y., Mir-Coll, J…
Journal: Nature Communications

Expanding the mass of pancreatic insulin-producing beta cells through re-activation of beta cell replication has been proposed as a therapy to prevent or delay the appearance of diabetes. Pancreatic beta cells exhibit an age-dependent decrease in their proliferative activity, partly related to changes in the systemic environment. Here we report...

Influenza Virus Neuraminidase Regulates Host CD8+ T-cell Response in Mice

Authors: Wu, C.Y., Chuang, H.Y., & Wong, C.H.
Journal: Communications Biology

Influenza A virus (IAV)-specific CD8+ T-cell response was shown to provide protection against pandemic and seasonal influenza infections. However, the response was often relatively weak and the mechanism was unclear. Here, we show that the composition of IAV released from infected cells is regulated by the neuraminidase (NA)...

Abnormally Activated OPN/integrin αVβ3/FAK Signalling is Responsible for EGFR-TKI Resistance in EGFR Mutant Non-Small-Cell Lung Cancer

Authors: Fu, Y., Zhang, Y., Lei, Z., Liu, T., Cai, T., Wang, A., Du, W., Zeng, Y…
Journal: Journal of Hematology & Oncology

Background: Acquired epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI) resistance limits the long-term clinical efficacy of tyrosine kinase-targeting drugs. Although most of the mechanisms of acquired EGFR-TKI resistance have been revealed, the mechanism of ~ 15% of cases has not yet been elucidated. Methods...

Hypusination Orchestrates the Antimicrobial Response of Macrophages

Authors: Gobert, A.P., Finley, J.L., Latour, Y.L., Asim, M., Smith, T.M…
Journal: Cell Reports

Innate responses of myeloid cells defend against pathogenic bacteria via inducible effectors. Deoxyhypusine synthase (DHPS) catalyzes the transfer of the N-moiety of spermidine to the lysine-50 residue of eukaryotic translation initiation factor 5A (EIF5A) to form the amino acid hypusine. Hypusinated EIF5A (EIF5AHyp) transports...

MiRNA‐516a Promotes Bladder Cancer Metastasis by Inhibiting MMP9 Protein Degradation via the AKT/FOXO3A/SMURF1 Axis

Authors: Chang, Y., Jin, H., Li, H., Ma, J., Zheng, Z., Sun, B., Lyu, Y., Lin, M…
Journal: Clinical and Translational Medicine

Background: Metastasis is the leading cause of death in patients with bladder cancer (BC). However, current available treatments exert little effects on metastatic BC. Moreover, traditional grading and staging have only a limited ability to identify metastatic BC. Accumulating evidence indicates that the aberrant expression of microRNA is...

Propofol Reduces Renal Ischemia/Reperfusion-induced Acute Lung Injury by Stimulating Sirtuin 1 and Inhibiting Pyroptosis

Authors: Liu, Z., Meng, Y., Miao, Y., Yu, L., & Yu, Q.
Journal: Aging

The activation of pyroptosis is an important feature of renal ischemia/reperfusion (rI/R)-induced acute lung injury (ALI). Propofol, a general anesthetic, is known to inhibit inflammation in I/R-induced ALI. We investigated whether propofol could suppress pyroptosis during rI/R-induced ALI by upregulating sirtuin 1 (SIRT1). We generated an...

Allicin Attenuated Advanced Oxidation Protein Product-Induced Oxidative Stress and Mitochondrial Apoptosis in Human Nucleus Pulposus Cells

Authors: Xiang, Q., Cheng, Z., Wang, J., Feng, X., Hua, W., Luo, R…
Journal: Oxidative Medicine and Cellular Longevity

Intervertebral disc degeneration (IDD) is one of the most common chronic degenerative musculoskeletal disorders. Oxidative stress-induced apoptosis of the nucleus pulposus (NP) cells plays a key role during IDD progression. Advanced oxidation protein products (AOPP), novel biomarkers of oxidative stress, have been reported to...
