Research Spotlight - August 2023

Welcome to this month's Research Spotlight where we delve into an array of exciting scientific discoveries!

For the field of neuroscience, scientists examined the significance of G3BP2-Tau interaction for Tau aggregation as well as the role of SMN for neurodegeneration. In gastroenterology, researchers presented modified prebiotic-based probiotics as a novel approach for treating colitis. Furthermore, another study offered insights into the connections between gut microbiota and diabetic polyneuropathy.

Turning to oncology, scientists investigated circular RNA variants in glioblastoma, which hold promise for diagnostics and treatment. In this issue, we also highlight discoveries like antioxidant mitoquinone, innovative extracellular vesicle-packaged ILK, α-tubulin acetylation's importance, and an artificial niche creation.

These advancements bring fresh strategies to address diverse challenges in different fields. Scroll down to find out more!

Increased G3BP2-Tau Interaction in Tauopathies is a Natural Defense Against Tau Aggregation

Authors: Wang, C., Terrigno, M., Li, J., Distler, T., Pandya, N.J., Ebeling, M…
Journal: Neuron

Many RNA-binding proteins (RBPs), particularly those associated with RNA granules, promote pathological protein aggregation in neurodegenerative diseases. Here, we demonstrate that G3BP2, a core component of stress granules, directly interacts with Tau and inhibits Tau aggregation. In the human brain, the interaction of G3BP2...

Modified Prebiotic-Based “Shield” Armed Probiotics with Enhanced Resistance of Gastrointestinal Stresses and Prolonged Intestinal Retention for Synergistic Alleviation of Colitis

Authors: Xie, A., Ji, H., Liu, Z., Wan, Y., Zhang, X., Xiong, H., Nie, S.P., Wan, H.
Journal: ACS Nano

Oral administration of probiotics is a promising method to alleviate inflammatory bowel diseases (IBDs). However, gastrointestinal environmental sensitivity and inferior intestinal colonization of probiotics hinder the alleviation effect. Here, we developed a simple yet effective modified prebiotic-based “shield” (Fe-TA@mGN) composed...

Gut Microbiota Modulate Distal Symmetric Polyneuropathy in Patients With Diabetes

Authors: Yang, J., Yang, X., Wu, G., Huang, F., Shi, X., Wei, W., Zhang, Y…
Journal: Cell Metabolism

The pathogenic mechanisms underlying distal symmetric polyneuropathy (DSPN), a common neuropathy in patients with diabetes mellitus (DM), are not fully understood. Here, we discover that the gut microbiota from patients with DSPN can induce a phenotype exhibiting more severe peripheral neuropathy in db/db mice. In a randomized...

Circular RNA encoded MET variant promotes glioblastoma tumorigenesis

Authors: Zhong, J., Wu, X., Gao, Y., Chen, J., Zhang, M., Zhou, H., Yang, J…
Journal: Nature Communications

Activated by its single ligand, hepatocyte growth factor (HGF), the receptor tyrosine kinase MET is pivotal in promoting glioblastoma (GBM) stem cell self-renewal, invasiveness and tumorigenicity. Nevertheless, HGF/MET-targeted therapy has shown limited clinical benefits in GBM patients, suggesting hidden mechanisms of MET signalling...

SMN Regulates GEMIN5 Expression and Acts As a Modifier of GEMIN5-Mediated Neurodegeneration

Authors: Fortuna, T.R., Kour, S., Chimata, A.V., Bühl, A.M., Anderson, E.N…
Journal: Acta Neuropathologica

GEMIN5 is essential for core assembly of small nuclear Ribonucleoproteins (snRNPs), the building blocks of spliceosome formation. Loss-of-function mutations in GEMIN5 lead to a neurodevelopmental syndrome among patients presenting with developmental delay, motor dysfunction, and cerebellar atrophy by perturbing SMN complex protein...

Extracellular Vesicle-Packaged ILK From Mesothelial Cells Promotes Fibroblast Activation in Peritoneal Fibrosis

Authors: Huang, Q., Sun, Y., Peng, L., Sun, J., Sha, Z., Lin, H., Li, Y., Li, C…
Journal: Journal of Extracellular Vesicles

Progressive peritoneal fibrosis and the loss of peritoneal function often emerged in patients undergoing long-term peritoneal dialysis (PD), resulting in PD therapy failure. Varieties of cell-cell communications among peritoneal cells play a significant role in peritoneal fibrogenesis. Extracellular vesicles (EVs) have been confirmed...

Authors: Jin, B.R., Lim, C.Y., Kim, H.J., Lee, M., An, H.J.
Journal: Redox Biology

Mitoquinone (MitoQ), a mitochondria-targeted antioxidant, has been used to treat several diseases. The present study aimed to investigate the therapeutic effects of MitoQ in benign prostatic hyperplasia (BPH) models and their underlying molecular mechanisms. In this study, we determined that MitoQ inhibited dihydrotestosterone (DHT)...
