Research Spotlight - August 2021

Our August edition of Research Spotlight presents 10 recent publications from scientists whom Boster has served.

In this month’s edition, research studies delve into the behavioral and cognitive parts of the brain and investigate how various cytokine levels are associated with the progression of neurodegenerative disorders. A comprehensive evaluation of the efficacy and safety of using stem cell therapy for traumatic brain injury is also discussed.

Researchers explore additional evidence to address major gaps in understanding the molecular mechanisms of stress, cancer and autoimmune diseases.

Astrocytic Interleukin-3 Programs Microglia and Limits Alzheimer’s Disease

Authors: McAlpine, C.S., Park, J., Griciuc, A., Kim, E., Choi, S.H., Iwamoto, Y…
Journal: Nature

Communication within the glial cell ecosystem is essential for neuronal and brain health. The influence of glial cells on the accumulation and clearance of β-amyloid (Aβ) and neurofibrillary tau in the brains of individuals with Alzheimer’s disease (AD) is poorly understood, despite growing awareness that these are therapeutically...

Pre-clinical Study of Human Umbilical Cord Mesenchymal Stem Cell Transplantation for the Treatment of Traumatic Brain Injury: Safety Evaluation from Immunogenic and Oncogenic Perspectives

Authors: Wang, G., Wu, H.L., Liu, Y.P., Yan, D.Q., Yuan, Z.L., Chen, L…
Journal: Neural Regeneration Research

>Stem cell therapy is a promising strategy for the treatment of traumatic brain injury (TBI). However, animal experiments are needed to evaluate safety; in particular, to examine the immunogenicity and tumorigenicity of human umbilical cord mesenchymal stem cells (huMSCs) before clinical application. In this study, huMSCs were...

CD147 Receptor is Essential for TFF3-Mediated Signaling Regulating Colorectal Cancer Progression

Authors: Cui, H.Y., Wang, S.J., Song, F., Cheng, X., Nan, G., Zhao, Y…
Journal: Signal Transduction and Targeted Therapy

Major gaps in understanding the molecular mechanisms of colorectal cancer (CRC) progression and intestinal mucosal repair have hampered therapeutic development for gastrointestinal disorders. Trefoil factor 3 (TFF3) has been reported to be involved in CRC progression and intestinal mucosal repair; however, how TFF3 drives tumors to...

An mTORC1-GRASP55 Signaling Axis Controls Unconventional Secretion to Reshape the Extracellular Proteome Upon Stress

Authors: Nüchel, J., Tauber, M., Nolte, J.L., Mörgelin, M., Türk, C., Eckes, B…
Journal: Molecular Cell

Cells communicate with their environment via surface proteins and secreted factors. Unconventional protein secretion (UPS) is an evolutionarily conserved process, via which distinct cargo proteins are secreted upon stress. Most UPS types depend upon the Golgi-associated GRASP55 protein. However, its regulation and biological role...

Bionic Microenvironment-Inspired Synergistic Effect of Anisotropic Micro-Nanocomposite Topology and Biology Cues on Peripheral Nerve Regeneration

Authors: Li, G., Zheng, T., Wu, L., Han, Q., Lei, Y., Xue, L., Zhang, L., Gu, X…
Journal: Science Advances

Anisotropic topographies and biological cues can simulate the regenerative microenvironment of nerve from physical and biological aspects, which show promising application in nerve regeneration. However, their synergetic influence on injured peripheral nerve is rarely reported. In the present study, we constructed a bionic...

Ceria Nanozyme-Integrated Microneedles Reshape the Perifollicular Microenvironment for Androgenetic Alopecia Treatment

Authors: Yuan, A., Xia, F., Bian, Q., Wu, H., Gu, Y., Wang, T., Wang, R…
Journal: ACS Nano

Androgenetic alopecia (AGA) is highly prevalent in current society but lacks effective treatments. The dysregulation of the hair follicle niche induced by excessive reactive oxygen species (ROS) and insufficient vascularization in the perifollicular microenvironment is the leading cause of AGA. Herein, we designed a ceria nanozyme...

Stromal Induction of BRD4 Phosphorylation Results in Chromatin Remodeling and BET inhibitor Resistance in Colorectal Cancer

Authors: Wang, W., Tang, Y.A., Xiao, Q., Lee, W.C., Cheng, B., Niu, Z…
Journal: Nature Communications

BRD4, a Bromodomain and Extraterminal (BET) protein family member, is a promising anti-cancer drug target. However, resistance to BET inhibitors targeting BRD4 is common in solid tumors. Here, we show that cancer-associated fibroblast (CAF)-activated stromal signaling, interleukin-6/8-JAK2, induces BRD4 phosphorylation at tyrosine...

Lactate and Glutamine Support NADPH Generation in Cancer Cells Under Glucose Deprived Conditions

Authors: Ying, M., You, D., Zhu, X., Cai., L., Zeng, S., Hu, X.
Journal: Redox Biology

Although glucose, through pentose phosphate pathway (PPP), is the main source to generate NADPH, solid tumors are often deprived of glucose, hence alternative metabolic pathways to maintain NADPH homeostasis in cancer cells are required. Here, we report that lactate and glutamine support NADPH production via isocitrate dehydrogenase...

Exosomes and Biomimetic Nanovesicles-Mediated Anti-Glioblastoma Therapy: A Head-to-Head Comparison

Authors: Wu, J.Y., Li, Y.J., Hu, X.B., Huang, S., Luo, S., Tang, T., Xiang, D.X.
Journal: Journal of Controlled Release

Exosomes (Exos) are promising vehicles for brain drug delivery due to nanosize and the ability to breach the blood–brain barrier (BBB). But the low yield of natural exosomes limits its application for nanomedicine. The generation of bioinspired nanovesicles (BNVs) that mimicking Exos is attractive, but there is a lack of comparative...
