Background of Wnt-2b
WNT2B (Wingless-Type MMTV Integration Site Family Member 2B), is a protein that in humans is encoded by the WNT2B gene. This gene encodes a member of the wingless-type MMTV integration site (WNT) family of highly conserved, secreted signaling factors. Katoh et al. (1996) used fluorescence in situ hybridization to map the WNT13 gene to chromosome 1p13. WNT family members function in a variety of developmental processes including regulation of cell growth and differentiation and are characterized by a WNT-core domain. This gene may play a role in human development as well as human carcinogenesis.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human HEK293 whole cell lysates,
Lane 2: human HEPG2 whole cell lysates,
Lane 3: human Jurkat whole cell lysates,
Lane 4: human A549 whole cell lysates,
Lane 5: human Raji whole cell lysates,
Lane 6: human SW620 whole cell lysates,
Lane 7: human SKOV3 whole cell lysates,
Lane 8: rat PC-12 whole cell lysates,
Lane 9: mouse Neuro-2a whole cell lysates,
Lane 10: mouse ANA-1 whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Wnt2b antigen affinity purified polyclonal antibody (Catalog # A04879-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Wnt2b at approximately 60KD. The expected band size for Wnt2b is at 60KD.
