Background of TRPC5

Short transient receptor potential channel 5 (TrpC5), also known as TRP-5, is a protein that in humans is encoded by the TRPC5 gene. TRPC5 is subtype of the TRPC family of mammalian transient receptor potential ion channels. It is mapped to Xq23. The predicted 973-amino acid TRPC5 protein has a calculated molecular mass of 111.5 kD. It contains the characteristic 8 predicted transmembrane domains (TM1 through TM8), including a pore region (TM7) between TM6 and TM8. TRPC5 is a multi-pass membrane protein and is thought to form a receptor-activated non-selective calcium permeant cation channel, and it is a candidate for the regulation of calcium waves. The protein is active alone or as a heteromultimeric assembly with TRPC1, TRPC3, and TRPC4.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Question

We are currently using anti-TRPC5 antibody PB9271 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on pig tissues as well?

Verified Customer

Verified customer

Asked: 2019-11-04

Answer

The anti-TRPC5 antibody (PB9271) has not been validated for cross reactivity specifically with pig tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in pig you can get your next antibody for free. Please contact me if I can help you with anything.

Question

Does PB9271 anti-TRPC5 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2018-12-26

Answer

You can see on the product datasheet, PB9271 anti-TRPC5 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question

Will anti-TRPC5 antibody PB9271 work on monkey WB with fetal brain?

K. Taylor

Verified customer

Asked: 2017-05-25

Answer

Our lab technicians have not tested anti-TRPC5 antibody PB9271 on monkey. You can run a BLAST between monkey and the immunogen sequence of anti-TRPC5 antibody PB9271 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated monkey samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in monkey fetal brain in WB, you can get your next antibody for free.
