Background of STI1
STIP1 is an adaptor protein that coordinates the functions of HSP70 and HSP90 in protein folding. It is thought to assist in the transfer of proteins from HSP70 to HSP90 by binding both HSP90 and substrate-bound HSP70. STIP1 also stimulates the ATPase activity of HSP70 and inhibits the ATPase activity of HSP90, suggesting that it regulates both the conformations and ATPase cycles of these chaperones. The International Radiation Hybrid Mapping Consortium mapped the STIP1 gene to 11q13.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human COLO320 whole cell lysates,
Lane 3: human MCF-7 whole cell lysates,
Lane 4: rat C6 whole cell lysates,
Lane 5: rat RH35 whole cell lysates,
Lane 6: mouse brain tissue lysates,
Lane 7: mouse liver tissue lysates,
Lane 8: mouse Neuro-2a whole cell lysates,
Lane 9: mouse HEPA1-6 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-STIP1 antigen affinity purified polyclonal antibody (Catalog # PB9896) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for STIP1 at approximately 63 kDa. The expected band size for STIP1 is at 63 kDa.

We are currently using anti-STIP1 antibody PB9896 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, rat. Is it likely that the antibody can work on horse tissues as well?

The anti-STIP1 antibody (PB9896) has not been validated for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity.
