Background of Rab18
Ras-related protein Rab-18 is a protein that in humans is encoded by the RAB18 gene. It is a ubiquitously expressed protein with particularly high expression in the brain. The protein encoded by this gene is a member of a family of Ras-related small GTPases that regulate membrane trafficking in organelles and transport vesicles. Knockdown studies in zebrafish suggest that this protein may have a role in eye and brain development. Mutations in this gene are associated with Warburg micro syndrome type 3. Alternatively spliced transcript variants have been found for this gene.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Testis Tissue Lysate at 50ug,
Lane 2: Rat Lung Tissue Lysate at 50ug,
Lane 3: 293T Whole Cell Lysate at 40ug,
Lane 4: HELA Whole Cell Lysate at 40ug,
Lane 5: HEPG2 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Rab18 antigen affinity purified polyclonal antibody (Catalog # PB9791) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Rab18 at approximately 23 kDa. The expected band size for Rab18 is at 23 kDa.

Question
We are currently using anti-Rab18 antibody PB9791 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, rat. Is it possible that the antibody can work on canine tissues as well?

The anti-Rab18 antibody (PB9791) has not been tested for cross reactivity specifically with canine tissues, though there is a good chance of cross reactivity.
