Background of Piccolo
Piccolo, also referred to as Aczonin, is a large protein which consists of an N-terminal Zn2+ finger, several piccolo-bassoon homology domains and C-terminal PDZ and C2 domains (1, 2). In general it is found together with bassoon, a related huge multi-domain protein of the CAZ (cytoskeletal matric assembled at active zones) (3). Piccolo is a scaffolding protein for proteins involved in endo- and exocytosis of synaptic vesicles. Piccolo has also been shown to interfere with clathrin medicated endocytosis by binding to the F-actin and dynamin binding protein Abp1 (2).

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Pclo antigen affinity purified polyclonal antibody (Catalog # M05881) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-Mouse IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # SA1021) with Tanon 5200 system. A specific band was detected for Pclo.

We are currently using anti-Piccolo Monoclonal antibody M05881 for rat tissue, and we are happy with the IF results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on pig tissues as well?
