Background of ALDH18A1
Delta-1-pyrroline-5-carboxylate synthetase (P5CS) is an enzyme that in humans is encoded by the ALDH18A1 gene. This gene is mapped to 10q24.1. This gene is a member of the aldehyde dehydrogenase family and encodes a bifunctional ATP- and NADPH-dependent mitochondrial enzyme with both gamma-glutamyl kinase and gamma-glutamyl phosphate reductase activities. The encoded protein catalyzes the reduction of glutamate to delta1-pyrroline-5-carboxylate, a critical step in the de novo biosynthesis of proline, ornithine and arginine. Mutations in this gene lead to hyperammonemia, hypoornithinemia, hypocitrullinemia, hypoargininemia and hypoprolinemia and may be associated with neurodegeneration, cataracts and connective tissue diseases. Alternatively spliced transcript variants, encoding different isoforms, have been described for this gene.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human MDA-MB-453 whole cell lysates,
Lane 3: human Caco-2 whole cell lysates,
Lane 4: human HEK293 whole cell lysates,
Lane 5: rat heart tissue lysates,
Lane 6: mouse skeletal muscle tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-ALDH18A1 antigen affinity purified polyclonal antibody (Catalog # A06119-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for ALDH18A1 at approximately 87KD. The expected band size for ALDH18A1 is at 87KD.

We are currently using anti-P5CS/ALDH18A1 antibody A06119-2 for rat tissue, and we are satisfied with the ELISA results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on dog tissues as well?

The anti-P5CS/ALDH18A1 antibody (A06119-2) has not been validated for cross reactivity specifically with dog tissues, though there is a good chance of cross reactivity.
