Background of Mad
MAD protein is a protein that in humans is encoded by the MXD1 gene. This gene encodes a member of the MYC/MAX/MAD network of basic helix-loop-helix leucine zipper transcription factors. The MYC/MAX/MAD transcription factors mediate cellular proliferation, differentiation and apoptosis. The encoded protein antagonizes MYC-mediated transcriptional activation of target genes by competing for the binding partner MAX and recruiting repressor complexes containing histone deacetylases. Mutations in this gene may play a role in acute leukemia, and the encoded protein is a potential tumor suppressor. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: rat testis tissue lysates,
Lane 2: mouse ovary tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Mad antigen affinity purified polyclonal antibody (Catalog # A06485-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Mad at approximately 28KD. The expected band size for Mad is at 25KD.

We are currently using anti-Mad/MXD1 antibody A06485-1 for rat tissue, and we are well pleased with the ELISA results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on zebrafish tissues as well?

The anti-Mad/MXD1 antibody (A06485-1) has not been validated for cross reactivity specifically with zebrafish tissues, but there is a good chance of cross reactivity.
