Background of PMEL17/SILV
By immunohistochemistry, it specifically recognizes a protein in melanocytes and melanomas. This monoclonal antibody reacts with junctional and blue nevus cells and variably with fetal and neonatal melanocytes. Intradermal nevi, normal adult melanocytes, and non-melanocytic cells are negative. It does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin.This monoclonal antibody labels formalin-fixed, paraffin-embedded melanomas and other tumors showing melanocytic differentiation.

Assay Dilutions Recommendation

Flow Cytometry (1-2ug/million cells)
Immunofluorescence (1-2ug/ml)
Immunohistochemistry (Formalin-fixed) (1-2ug/ml for 30 minutes at RT)(Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at RT for 20 minutes)

Our team were well pleased with the WB result of your anti-gp100 / Melanosome / PMEL17 / SILV (Melanoma Marker) Monoclonal antibody. However we have seen positive staining in placenta endoplasmic reticulum membrane using this antibody. Is that expected? Could you tell me where is PMEL supposed to be expressed?

According to literature, placenta does express PMEL. Generally PMEL expresses in endoplasmic reticulum membrane. Regarding which tissues have PMEL expression, here are a few articles citing expression in various tissues:

From what I have seen in literature melanoma does express PMEL. From what I have seen in Uniprot.org, PMEL is expressed in pigmented layer of retina, placenta spleen, placenta, melanoma, among other tissues. Regarding which tissues have PMEL expression, here are a few articles citing expression in various tissues:

We are currently using anti-gp100 / Melanosome / PMEL17 / SILV (Melanoma Marker) Monoclonal antibody M19774 for human tissue, and we are satisfied with the IHC results. The species of reactivity given in the datasheet says equine, human. Is it possible that the antibody can work on zebrafish tissues as well?

The anti-gp100 / Melanosome / PMEL17 / SILV (Melanoma Marker) Monoclonal antibody (M19774) has not been validated for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity.
