Background of FOXC1
Forkhead box C1, also known as FOXC1, is a protein which in humans is encoded by the FOXC1 gene. It is mapped to 6p25.3. This gene belongs to the forkhead family of transcription factors which is characterized by a distinct DNA-binding forkhead domain. The specific function of this gene has not yet been determined; however, it has been shown to play a role in the regulation of embryonic and ocular development. Mutations in this gene cause various glaucoma phenotypes including primary congenital glaucoma, autosomal dominant iridogoniodysgenesis anomaly, and Axenfeld-Rieger anomaly.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human HEK293 whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-FOXC1 antigen affinity purified polyclonal antibody (Catalog # A00962-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for FOXC1 at approximately 75KD. The expected band size for FOXC1 is at 57KD.

We are currently using anti-FOXC1 antibody A00962-2 for human tissue, and we are satisfied with the ELISA results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on feline tissues as well?

The anti-FOXC1 antibody (A00962-2) has not been tested for cross reactivity specifically with feline tissues, but there is a good chance of cross reactivity.
