Background of FGF-22
FGF22, Fibroblast growth factor 22, is a protein which in humans is encoded by the FGF22 gene. The FGF22 sequence in a genomic clone is assigned to 19p13.3. The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. In situ hybridization demonstrated expression in the inner root sheath of the hair follicle. FGF22 is involved in hair development.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human K562 whole cell lysates,
Lane 2: human Caco-2 whole cell lysates,
Lane 3: human U-87MG whole cell lysates,
Lane 4: human PC-3 whole cell lysates,
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-FGF22 antigen affinity purified polyclonal antibody (Catalog # PA1455) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for FGF22 at approximately 24KD. The expected band size for FGF22 is at 20KD.

We are currently using anti-FGF22 antibody PA1455 for rat tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on zebrafish tissues as well?

The anti-FGF22 antibody (PA1455) has not been validated for cross reactivity specifically with zebrafish tissues, but there is a good chance of cross reactivity.
