Background of EAAT3
Solute carrier family 1 member 1, also called SLC1A1, is a protein that in humans is encoded by the SLC1A1 gene. By Southern analysis of a panel of human/rodent somatic cell hybrids and by fluorescence in situ hybridization (FISH), this gene is mapped to 9p24.2. This gene encodes a member of the high-affinity glutamate transporters that play an essential role in transporting glutamate across plasma membranes. In brain, these transporters are crucial in terminating the postsynaptic action of the neurotransmitter glutamate, and in maintaining extracellular glutamate concentrations below neurotoxic levels. This transporter also transports aspartate, and mutations in this gene are though to cause dicarboxylicamino aciduria, also known as glutamate-aspartate transport defect.

Will anti-EAAT3/SLC1A1 antibody PA2164-2 work on feline WB with brain cortex?

Our lab technicians have not validated anti-EAAT3/SLC1A1 antibody PA2164-2 on feline. You can run a BLAST between feline and the immunogen sequence of anti-EAAT3/SLC1A1 antibody PA2164-2 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated feline samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in feline brain cortex in WB, you can get your next antibody for free.

We are currently using anti-EAAT3/SLC1A1 antibody PA2164-2 for mouse tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says mouse, rat. Is it likely that the antibody can work on zebrafish tissues as well?

The anti-EAAT3/SLC1A1 antibody (PA2164-2) has not been validated for cross reactivity specifically with zebrafish tissues, but there is a good chance of cross reactivity. Answer
As indicated on the product datasheet, PA2164-2 anti-EAAT3/SLC1A1 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
