Background of Desmoglein-3
Desmoglein3 is also known as PVA (Pemphigus Vulgaris Antigen). Pemphigus vulgaris (PV) and pemphigus foliaceus (PF) are autoimmune diseases of the skin which have as target antigens 2 different members of the desmoglein subfamily of the desmosomal cadherins: pemphigus vulgaris antigen (PVA, or desmoglein-3) in the case of PV and desmoglein I in the case of PF. The DSG3 gene, like the genes for DSG1 and DSG2, is located on chromosome 18. The human DSG3 gene consists of 15 exons and spans more than 23 kb. DSG3 and DSG1 are the autoantigens of pemphigus vulgaris and pemphigus foliaceus, respectively. DSG3 and DSG1 are the target antigens in PNP, and IgG autoantibodies against DSG3 in PNP sera play a pathogenic role in the loss of keratinocyte adhesion and blistering.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human A431 whole cell lysates,
Lane 2: human Hacat whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-DSG3 antigen affinity purified polyclonal antibody (Catalog # PA1567) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for DSG3 at approximately 130 kDa. The expected band size for DSG3 is at 107 kDa.

According to literature tongue does express DSG3. According to Uniprot.org, DSG3 is expressed in mouth mucosa, tongue, saliva, among other tissues. Regarding which tissues have DSG3 expression, here are a few articles citing expression in various tissues:
