Background of CEBP Delta

This antibody is suitable for Cancer, Immunology and Nuclear Signaling research. C/EBP proteins are DNA-binding proteins that are important transcriptional activators in the regulation of genes involved in immune and inflammatory responses and may play an important role in the regulation of the several genes associated with activation and/or differentiation of macrophages. Members of the C/EBP transcription factor family are related by a high degree of amino acid sequence identity to the basic leucine zipper DNA-binding domain and show distinct but overlapping patterns of tissue- and stage-restricted expression. Osada et al. identified the consensus binding site for the family as RTTGCGYAAY (R = A or G, and Y = C or T). Phosphorylation of C/EBPd may increase binding activity, whereas phosphorylation of the a and b family members may decrease binding affinity. C/EBPd is a nuclear protein that binds DNA as a dimer and can form stable heterodimers with all other C/EBP family members.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CEBPD antigen affinity purified polyclonal antibody (Catalog # A03499-2) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-Rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # SA1022) with Tanon 5200 system. A specific band was detected for CEBPD.
