Background of CDC6
Cell division control ptotein 6 homolog, also called CDC18L or CDC6, is a protein that in humans is encoded by the CDC6 gene. By analysis of somatic cell hybrids and by fluorescence in situ hybridization, this gene is mapped to 17q21.2. The protein encoded by this gene is highly similar to Saccharomyces cerevisiae Cdc6, a protein essential for the initiation of DNA replication. And this protein functions as a regulator at the early steps of DNA replication. It localizes in cell nucleus during cell cyle G1, but translocates to the cytoplasm at the start of S phase. The subcellular translocation of this protein during cell cyle is regulated through its phosphorylation by Cdks. Transcription of this protein was reported to be regulated in response to mitogenic signals through transcriptional control mechanism involving E2F proteins. This gene is involved in the initiation of DNA replication.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Brain Tissue Lysate at 50ug,
Lane 2: JURKAT Whole Cell Lysate at 40ug,
Lane 3: MM231 Whole Cell Lysate at 40ug,
Lane 4: HT1080 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Cdc6 antigen affinity purified polyclonal antibody (Catalog # PB9531) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Cdc6 at approximately 63 kDa. The expected band size for Cdc6 is at 63 kDa.

Cdc6 was detected in a paraffin-embedded section of rat testis tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1 μg/ml rabbit anti-Cdc6 Antibody (PB9531) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB as the chromogen.

We are currently using anti-Cdc6 antibody PB9531 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human, rat. Is it true that the antibody can work on zebrafish tissues as well?

The anti-Cdc6 antibody (PB9531) has not been tested for cross reactivity specifically with zebrafish tissues, but there is a good chance of cross reactivity.
