Background of 4-1BB/TNFRSF9/CD137
CD137 is a member of the tumor necrosis factor (TNF) receptor family. This receptor contributes to the clonal expansion, survival, and development of T cells. It can also induce proliferation in peripheral monocytes, enhance T cell apoptosis induced by TCR/CD3 triggered activation, and regulate CD28 co-stimulation to promote Th1 cell responses. The expression of this receptor is induced by lymphocyte activation. TRAF adaptor proteins have been shown to bind to this receptor and transduce the signals leading to activation of NF-kappaB.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: recombinant mouse CD137 protein 1ng.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD137 antigen affinity purified polyclonal antibody (Catalog # A00867-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CD137 at approximately 65KD. The expected band size for CD137 is at 44KD.

We are currently using anti-CD137/Tnfrsf9 antibody A00867-2 for rat tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says mouse, rat. Is it likely that the antibody can work on monkey tissues as well?

The anti-CD137/Tnfrsf9 antibody (A00867-2) has not been tested for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity.
